Therapeutic Potential of Tolerogenic Dendritic Cells in IBD: From Animal Models to Clinical Application
نویسندگان
چکیده
The gut mucosa undergoes continuous antigenic exposure from food antigens, commensal flora derived ligands, and pathogens. This constant stimulation results in controlled inflammatory responses that are effectively suppressed by multiple factors. This tight regulation, necessary to maintain intestinal homeostasis, is affected during inflammatory bowel diseases (IBD) resulting in altered immune responses to harmless microorganisms. Dendritic cells (DCs) are sentinels of immunity, located in peripheral and lymphoid tissues, which are essential for homeostasis of T cell-dependent immune responses. The expression of a particular set of pathogen recognition receptors allows DCs to initiate immune responses. However, in the absence of danger signals, different DC subsets can induce active tolerance by inducing regulatory T cells (Treg), inhibiting inflammatory T helper cell responses, or both. Interestingly, several protocols to generate clinical grade tolerogenic DC (tol-DCs) in vitro have been described, opening the possibility to restore the intestinal homeostasis to bacterial flora by cellular therapy. In this review, we discuss different DC subsets and their role in IBD. Additionally, we will review preclinical studies performed in animal models while describing recent characterization of tol-DCs from Crohn's disease patients for clinical application.

متن کاملEvaluation of the Effect of the 47 kDa Protein Isolated from Aged Garlic Extract on Dendritic Cells
Objective(s) Garlic (Allium sativum) is known as a potent spice and a medicine with broad therapeutic properties ranging from antibacterial to anticancer, and anticoagulant. One of the major purified garlic protein components is the 47 kDa protein. In this study, the effect of 47 kDa protein extracted from aged garlic (AGE) was evaluated on mouse dendritic cell (DC) maturation in vitro. Mate...
متن کاملCD40 Knocked Down Tolerogenic Dendritic Cells Decrease Diabetic Injury
Background: Type-1 diabetes (T1D) is an autoimmune disease in which T lymphocytes destroy insulin-producing β-cells. Control of self-reactive T lymphocytes and recovery of diabetic injury is the end point of T1D. Objective: To investigate generation of tolerogenic dendritic cells (tolDCs) as an innovative method of diabetes therapy. Methods: Lentivirus vector production was achieved by GIPZ mou...
متن کاملPotential Vaccines for Treating Crohn\'s Disease
Crohn's disease (CD) is an inflammatory disease of the gastrointestinal tract (GIT) tract and can affect several parts of the digestive system. There is a relationship between impaired mucosal barrier in the GIT of inflammatory bowel disease patients and the role of bacteria such as Mycobacterium avium paratuberculosis in CD. Apart from different therapeutic approaches for treating CD, developm...
متن کاملBone marrow dendritic cells deficient for CD40 and IL-23p19 are tolerogenic in vitro
Objective(s): In addition to pro-inflammatory role, dendritic cells (DCs) can also be anti-inflammatory when they acquire tolerogenic phenotype. In this study we tested the role of CD40 and IL-23p19 in antigen presenting function of bone marrow-derived DCs (BMDCs) by comparing BMDCs derived from CD40 knockout (CD40KO-DCs) and IL-23p19 (IL-23p19KO-DCs) knockout mice wit...
